چکیده

The aim of the present study reported here is to examine the production, purification and characterization of the enzyme laccase from the mushroom Hypsizygus ulmarius. The culture conditions were optimized for the maximum production of enzymes with different parameters such as incubation time, carbon, nitrogen sources. The enzyme was purified by ammonium sulphate precipitation and partially characterized. The optimum conditions for the maximum production of enzyme were; incubation period 6 day, pH 5.0, temperature 30C, inoculum 3%, carbon source (xylose 3%) and nitrogen source (KNO3 3%). The enzyme was purified to 5.11 fold with the recovery of 53.33%. The optimum pH and temperature for the purified laccase was found to be 6.0 and 40C respectively. The enzyme activity was enhanced by the metal ions Mnand Cu whereas it was reduced by Fe, Na and Co. These results suggest that Hypsizygus ulmarius could produce high yield of laccase. With its distinct characteristics laccase could be used for various applications.

Studies on the bioactive compounds of selected edible basidiomycetes mushroom fungi

The present study evaluated the anti-cell proliferation properties of edible basidiomycetes mushroom fungi against human breast cancer MCF-7 cell lines. The cells were treated with Dichloromethane (DCM) mycelial extracts of the mushroom fungi at different concentrations. Cytotoxic studies on the breast cancer cell lines (MCF -7) showed the anti-cell proliferation activity of DCM mycelial extrac...

Phenol Tolerance and Degradation Profile of Novel Edible Mushroom Hypsizygus Ulmarius in Ligninolytic and Non- Ligninolytic Media

Phenol tolerance and degradation using spent mycelium substrate of Hypsizygus ulmarius showed that ligninolytic media with carbon or nitrogen limitation gave highest growth (26.27sq.cm), while limitation of both nutrients reduced it to 8.1sq.cm. 7.68sq.cm was seen in non-ligninolytic media; simultaneous nutrient increase gave 1.16sq.cm. Tolerance to phenol was best in ligninolytic media with ca...

Purification and characterization of a novel laccase from the mushroom Pleurotus nebrodensis.

A novel laccase with a molecular mass of 64 kDa and the N-terminal sequence AIGPDDTINF was isolated from fresh fruiting bodies of the mushroom Pleurotus nebrodensis. The purification protocol comprised ion exchange chromatography on DEAE-cellulose, CM-cellulose, and Q-Sepharose, and gel filtration on Superdex 75. The laccase was adsorbed on DEAE-cellulose and Q-Sepharose, but not on CM-cellulos...
